Usually a person who is planning on taking a car on an extended road trip wants to do preventive maintenance and a safety inspection.
The inspection would include things like checking the tires for sufficient tread, inflation, and looking to see if their are any cuts on the sides or valve stem from coming too close to the curb. Making sure that there isn't excessive play in the steering wheel. That the brakes stop evenly, the peddle doesn't go down too far, that the emergency brake will keep the car stationary with it in drive. Checking all fluid levels under the hood and looking for leaks there and under the vehicle. Checking the battery connections for a tight fit and corrosion. Making sure the different belts under the hood are tight and don't look cracked. Making sure there is nothing loose under the hood or under the vehicle. Wiper blades, all lights including 4 ways. Safety equipment like a spare tire, flares or reflectors, or first aid kit, or fire extinguisher.
For the preventive maintenance, first taking care of any items that the safety inspection showed needing correction and then bringing whatever periodic maintenance the vehicle manufacturer recommends up to date, like oil changes, tune ups, transmission fluid & filter, etc..

Make sure the spare tire is filled up with air and has no holes (and that it's actually there).
Check for jack and changing supplies (i.e., a tire pump)
Change the oil, oil filter and air filter (if the trip is especially long)
Check air pressure on tires (to get better mileage)
Rotate the tires if they haven't been rotated in a while
Put a blanket, water, flashlight (with fresh batteries) and first aid kit in the car (just in case)
Take a cell phone
